有氧运动和二甲双胍对2型糖尿病大鼠血管炎症的影响及机制

Effects of Aerobic Exercise and Metformin on Vascular Inflammation in Type 2 Diabetic Rats and Its Mechanism

      目的  探讨有氧运动和二甲双胍对2型糖尿病大鼠血管炎症的影响及机制。
      方法  对2型糖尿病大鼠分别进行8周有氧运动,二甲双胍与二甲双胍+运动联合干预,检测干预后2型糖尿病大鼠糖脂代谢、主动脉形态、血管炎症标志物、氧化应激和NF-κB信号相关指标。
      结果  糖尿病对照组血清IL-1β,TNF-α、MCP-1和VCAM-1浓度均显著高于正常对照组,有氧运动及联合干预能显著降低大鼠血清IL-1β、TNF-α、MCP-1和VCAM-1浓度;有氧运动及联合干预能够改善2型糖尿病大鼠血糖代谢,降低血管氧化应激,下调NF-κBp65蛋白和mRNA表达,上调IκBα蛋白表达。
      结论  有氧运动与联合干预可能部分通过改善2型糖尿病大鼠血糖代谢,降低血管氧化应激,抑制NF-κB信号缓解2型糖尿病大鼠血管炎症。

     

    Abstract:
      Objective  It is to evaluate the effects of aerobic exercise and metformin on vascular inflammatory reaction in type 2 diabetic rats and explore its mechanism.
      Methods  Type 2 diabetic rates were treated with aerobic exercise, metformin and their combination therapies respectively for 8 weeks.After receiving intervention treatment, type 2 diabetic rats were tested to obtain relevant indicators such as glycolipid metabolism, aorta morphology, vascular inflammatory markers, oxidative stress, and NF-κB signal.
      Results  Serum concentrations of IL-1β, TNF-α, MCP-1 and VCAM-1 in DM group were significantly higher than those in Con group.Aerobic exercise and the combination of aerobic exercise and metformin could greatly decrease serum concentrations of IL-1β, TNF-α, MCP-1 and VCAM-1, improve glycolipid metabolism, relieve oxidative stress, down-regulate protein and mRNA express of NF-κBp65 and up-regulate express of IκBαin type 2 diabetes rats.
      Conclusions  Both aerobic exercise and the combination of aerobic exercise and metformin can effectively relieve the vascular inflammation in type 2 diabetic rats partly through the improvement of glucose metabolism, reduction of vascular oxidative stress, and inhibition of NF-ΚB signal.
